Duty officers of all qualities - up to and including purple - are currently planned to be accessible both via chance in certain assignments as well as rewards for achieving commendation tiers.
Random packs will also be available via the C-Store. The current plan is for some rare duty officers to be available only in game and some to be available only through random C-Store packs.
Note, however, that with the plan of allowing players to exchange dilithium (the working name for the new Emblem-like currency) for C-Store points, even the C-Store packs will be available to players through this exchange mechanism, if so desired.

Could we have some way of modifying the portrait of the DOff?
I'm not looking at anything fancy, just something colorful to help organization. With projected DOff capacities of triple digits, I'd like to be able to add some visual cues to the portrait.
For example, just a simple colored stripe from the upper right corner, down to the lower left corner that we can choose the color.
What got me thinking about this is the Unscrupulous and Scrupulous traits, and I just expanded on it.
For me, it would be black stripe for Unscrupulous (my "black ops" team),
Orange stripe for any I am willing to discard (my "red shirt" brigade),
White for the Scrupulous ones,
Etc..
We currently know skills like GW, Boarding Part, Tractor/Tachyon Beam and probably some I can't remember all can be improved via Doffs. So without asking for any detail information on the improvements themselves... can we improve all our BO/captain skills with Doff traits?
Not all of them for launch. Eventually, I would like to see that, but there are a lot of interlocking pieces.
One question: Earlier it was stated that a ship would have to be in a given world's respective sector if an assignment required it, with the given example being the recruitment of an Andorian officer from Andoria. Does this mean we will finally get to know where the new Orion capital, Ter'jas Mor, is located? And how will the sort-of-vagrant Nausicaans be handled? Or will the KDF be "limited" to getting everything and anything from Qo'noS?

In the case of species that do have a system and social zone, we've also tried to make sure appropriate recruitment assignments can be gained from there in addition, since it would be kind of weird to be able to get Andorian recruitment assignments in the Sirius sector block but not in actual Andorian system space.
So, in the case of the Orion capital, a system has not been built for it, but the sector is there. Gorn were obviously a little easier, but only a little. The main issue is the bulk of the Klingon Empire and its vassal states lie outside the currently playable map.

Characters, whether existing or new characters who haven't bothered to open the system, will be given a stock set of duty officers based on your character's rank. The stock set is intended to allow you to start playing with the system and to start filling out your active roster, but will be basic.

We've tried to include assignments themed to as many areas as possible, including Orions. I cannot promise that every species will start with assignments, but I am expanding the breadth and depth of available assignments as fast as I can.
The screenshots are amazing, I just got around to seeing them all now. I just can't wait, I haven't played a lot lately some of it because of the content drought... but mainly because I have a habit of going 'dark' a month or two before a season release, followed by heavy play on tribble during testing, followed by all out war when the season hit's holodeck. The doff system is sort of giving me the same type of anticipation.
I'm not one to talk a lot about stuff on the forums as people have the same or better thoughts than I have, but seeing the screenshots.. wow! The missions, the abilities, great stuff. To me this is what Star Trek is. Of course you get to do cool stuff and fight in battles, but in the beginning of just about every episode it starts with the Captain entering into his/her log about doing the very stuff there that is listed as the doff missions. It will be fun to manage and take on that aspect of a star ship Captain. I'm glad you were able to run with this and it turned into what it is now.

Regular missions: Take the above themes and make something Andorian or Orion themed.

A ghost ship mission. For example, some famous ship, Say the Enteprise C or some such, that was presumed destroyed or mysteriously vanished 20+ years ago, has been detected on long range scans. Its your job to investigate it. Since you've no idea what's going to happen, it would be a multi-Doff assignment; at least 3 so you can have 1 of each profession investigating it. You guys can determine the rewards and stuff (ghost ship missions are supposed to be a mystery after all).
Actually, you know what? That would also make a good actual mission.

A joined trill member of your crew wants your help giving her or his close colleagues on board your ship some extra leave time that would allow them to assist the trill in question with her/his zhian'tara ritual.
For me it's definitely archaeology.
I liked Picard's interest with the T'Kon and with the power struggles as they are right now.. finding rare ancient artefacts could be very cool.

Morally questionable ones. Ones where we're probably (if not definitely) sending our DOFF(s) to their deaths (even if the mission is successful), but it's for a great cause like a key piece of intelligence or saving a life/ship (or shinies!). A bit like Spock's sacrifice in Wrath of Khan, Sito's death in Lower Decks, or Data's death in Nemesis. Ones where you have to take it on yourself to decide whether a single life is more important than another, several, or many.
Or even ones where we must kill innocents to protect the greater good; maybe there's some amazing slipstream technology that would allow the Borg to just appear at Earth/Q'onos without warning, on a research station that they're attacking to get the tech., and theonly way to stop the Borg from getting it is to destroy the station and all on it.

If your Duty Officers successfully retrieve your dog, it appears on your bridge, playing or sleeping or what have you.
