I wish i knew more of the lore and places of the Forgotten Realms campaign, but sadly I don't So I'd have to go with Neverwinter on account of it has the makings of a the type of city a lone adventurer type like i tend to RP as would travel to and from for taking on jobs and gathering supplies for my adventures.
I never played a pen and paper D&D game session when starting from anywhere unless you count Living Forgotten Realms games. I would have to say though if you did, Cormyr by far is nice with Moonshale Island being an interesting second (despite being so fricken dangerous if you aren't built for water combat). Though Kara-tur could be interesting if I played a game that went over that way and knew more about it.

Zhentil Keep! (That counts as a city, right?) Ran some delicious infiltration-style campaigns there against the Zhentarim battling their efforts around the Moonsea, which was always my favourite area to set stories in.

i've always prefered campaigns that started in the smaller cities, or towns. from the smaller places there is more of a sense of adventure hearing stories of the hero's having a bard come to town telling a story of Drizzt...makes for more of a story if perhaps you end up in those "big" cities and hear stories/meet the people you heard story's of as a new adventurer.

As far as PC games go, Neverwinter wins. For PnP I've got to say Myth Drannor as it was a long running campaign (years) that many characters from our group participated in various ways to restore the city.
