Try the "Verify" button in the start screen, this should find any broken files and fix your problem.

Maybe you've already taken another mission from the Gem Merchant, which requires you to exchange Earth, Heaven and Human Stone, 3 of each, for a Mirage Stone. This will block the 25 Celestones mission. If so, trash this 3*3 mission, and you will be able to proceed.

It depends more on personal preferences than on level. If you don't like to take a rest while waiting for your HP to recover, e.g. when farming mobs, just implant your HP charm and you don't need to pay much attention to your HP bar.
