The two characters I absolutely WILL be making are Halfling Greatweapon Fighter and Tiefling Devoted Cleric. I like to play atypical characters and both of these are just great for that. I would also like to get a Drow trickster rogue(or dwarf warlock if what I've been reading comes true) if I have the money to spare at launch.
I would go with the Dwarf Warlock as it ties directly into my first 4E character who's backstory was that he turned to the easy power of warlocks to hunt the demons who killed his parents when he was young. However he got all of that business sorted and out of the way by the time he was 30 and after 250 years of a pleasant marriage and a fulfilling fatherhood over his twin sons an old friend called him out of "retirement" to pick up the trade again.
Edit: I'm so surprised that most people haven't put a race down as well as a class.
Devoted Cleric, I have always loved being a healer in MMOs and even while playing D&D 4th edition I have centered my Cleric strictly to keep the group alive. It's just the thrill of knowing that you can change the balance of a battle by just being there and doing your job.

Definitely the devoted cleric for me, unless druids happen to be released by then (alas, that won't happen)
