Duplicate the quest - wipe-out everything except the items, NPCs and other things you want to keep. Start building-out from there. I actually keep a "template" quest (never published) that I create my NPCs and Items on and always duplicate that quest to start building a new one.
Like Angry Sprite said! You can also do this if you have a "home base" map like I and others do.

I have taken your advise and am now doing this. I used some of my Act I stuff to build Act 3.
The problem now is I added and built a cool map in Act 3 and I would love to go back and add it to Act 1, without having to rebuild the whole thing. But I take it there is no way around this, at least at the moment.

I keep a single "template" quest where I build all my items and reusable NPCs there. This way I just duplicate that and everything's in place.
When I want to create a new NPC I do it in the template first, take basic notes then immediately recreate it in the current in-progress quest. That's the best answer I have. It would be great if there were a "user library" where your created assets could be copied into for some ease with repeatable NPCs and items across quests, but at this point that is a pipe dream.
