I tried...
a. Running as administrator.
b. Adding the entire game directory to Comodo's trusted file list.
c. Turning off Comodo all together
d. Restarting my PC
e. Making all files writable
Always the same error. My log looks like this...

Previous patches (up until now) always worked with no problems.

same here, got the same massage like radnok,
i allready send a ticket to support because of the AH/AD exploit... now i cant even start the game anymore since 5 days...i just want my 189.99€ back!
I was thinking of getting the founders pack then this happened, glad I didn't get it first! Good luck getting your money back ripkordinal. I don't think I could now part with any money knowing that this game could lock me out at any time and there'll be no help overcoming the problem. Be warned people!
For players with a "The patchclient got a message while it was in a state with no response function" error, please download this file and replace your Neverwinter.exe file with this where you installed the game.
That's not the error I was getting. BUT overwriting the EXE with the one provided in the link did fix my problem. So anyone else that is getting 'Error 36 file locked' messages when patching (with similar logs to my previous posts above) might want to try it as well!

To be short just replace your Neverwinter.exe with this:
http://download.perfectworld.com/nw/launcher/Neverwinter.exe (This file is from the developers)
