I logged in and played about 2 hours last night with a new Romulan character. Do I need to do the same thing with my Federation and KDF character to get the test reward?
No. You simply need to play 1 hour to get your free tribble.
STO is about my Liberated Borg Federation Captain with his Breen 1st Officer, Jem'Hadar Tactical Officer, Liberated Borg Engineering Officer, Android Ops Officer, Photonic Science Officer, Gorn Science Officer, and Reman Medical Officer jumping into their Jem'Hadar Carrier and flying off to do missions for the new Romulan Empire. But for some players allowing a T5 Connie to be used breaks the canon in the game.
A weapon, I doubt. I don't think they've ever given away a weapon for testing. They did give a Doff at one point, though, so I wouldn't be surprised. Especially since there's already a Romulan Tribble.

I really hope it isn't another tribble. I barely have from for all the other tribbles and pets I currently have. A purple doff would be nice I suppose.

Based on previous testing rewards, I would say a Tribble is all but certain. And yes, it will most likely give out one Nukara Mark every hour, like ever other test reward tribble.
How do we know we've done the reward time? (and please don't say time yourself - just something like, to confirm we have done so).
I've played on tribble this weekend for well over an hour, and submitted a few bugs in the process.
I was wondering if anything pops up on screen to tell you? Like a "Thank you" message? If so, I think I have missed the message. If there isn't one, then oh well. I know I've done more than an hour! XD

If Cryptic actually gave a time, or a thank you message, then everyone would quit testing at that point... Your testing time is completely voluntary. If you want to stick around on Tribble and explore the new offerings, then that is your decision.
(Though the Dev's have been hinting that there might be additional rewards for 'serious' testers, what ever that means...)
Season 3 gave the Federation Runabout and the Klingon Empire Toron Shuttle pet for their Tribble Test Weekend reward and Season 6 gave a purple duty officer as well as the Fleet tribble so it is possible to get something besides a Nukara Tribble.
If you want to check to see if you qualified, then type /played since they wiped all characters and only new characters could be played for this weekend.

The doffs they gave in season 5 and 6 were tied to special conditions - season 5 doffs were tied to gaining doff commendations and the season 6 one was for doing missions on Nukara. There's no special objective given for LOR.
When they have a simple play-for-an-hour test weekend, it's always been a tribble except for season 3, which was the runabout noncombat pet.
Season 1, 2, and 4-7 all had a tribble themed to the season tied to the 1-hour condition. 5-7 were all tribbles that gave a resource introduced in that season - dilithium, fleet marks, and Romulan marks. Nukara marks are new this season, so smart bets are on the boring option of a Nukara mark tribble.
An oddball possibility is an EV suit. Tholians on the ground require an EV suit either on the player or the Tholians, and the new enemy faction, the Elachi, also benefits from an EV suit. It's not required, but when fighting them on their own ships, your character will periodically cough on fumes if he's not wearing an EV suit.
