No, you can go to both places as soon as you're done with the tutorial. However, there wont be anything to do on DS9 until around level 40, and Risa doesnt have anything other than during the Summer event.

There are no stupid questions... just stupid people...
Yeah, nothing specifically to do on DS9, but it has:
- Bank Inventory / Mail / Exchange consoles
- Vendors to sell your junk
- A "shipyard" area
- Quark's where you can play Dabo, or talk to Morn until your ears fall off
- A Ferengi with some Doff trade missions
- Security Officers to turn in Contraband for Dilithium
You can eventually transwarp directly to DS9, but you will need to max out your Diplomacy / Marauding commendation to Tier 4 by doing Doff (duty officer) missions in those categories. See link below for general info.
Also, here's a link to the story missions in STO if you are interested. There are plenty of missions that are worth repeating to collect rewards (though you may tear your hair out after a while) such as the current Featured Episode; Mirrors and Smoke.
Well, the story missions are mostly designed to be pretty easy though once you get into the missions involving the Romulans you will be challenged. Romulans are meant to be a bit overpowered in space combat so don't be too surprised if you die when going up against them. The best advice I can give specifically for them are:
1. Stay at least 5km away from them otherwise they will tractor beam you.
2. Have Beam Fire At Will / Cannon Scatter Volley ready to destroy the high yield plasma torpedoes they will be launching at you.
3. Polarized Hull (science Boff [bridge officer] ability can be used to temporarily negate the affects of tractor beams.
4. Evasive Maneuvers can also help you get away when you have a tractor beam on your ship. If you have two tractor beams on you then you move much slower. You are dead in the water if you have 3 tractor beams on you though.
5. Hazard Emitter (science Boff) can be used to stop any DoT (damage over time) effects like plasma weapon burns. It also heal hull damage over time.
Regardless what career / ship you command I generally recommend you have the following Boff abilities; assuming the ship you have allows you to slot them. Generally speaking, by the time you get your Tier 3 starship you should be able to slot in all of the abilities. A link to a description for all Boff abilities in space and on ground is provided below:
1. Engineering Team - Heals ship hull.
2. Emergency Power To Shields - Heals shields and shield damage resistance.
3. Hazard Emitter - Mentioned above and also hull damage resistance.
4. Science Team - Heals shields.
As mentioned in my previous post, there are missions that are worth repeating to get some good free gear to get you through the game. For some time I have been focusing more on ground combat than space combat in STO. Ground combat is clunky compared to more modern 3rd person shooter games, but I have grown to like to "old school" style in STO. Having said that, "The Core of the Matter" rewards the Neutronium Laced Combat Armor which is a pretty good early reward for you and your Boffs. It can be upgraded to Mk XII for free which means it is a good armor to use as you rank up your character. The mission is part of the Yesterday's War story arc which was introduced with the release of Agents of Yesterday (AoY).
As for ship gear... well the missions gives you what you need you can repeat them to collect multiple gear and higher mark levels when you rank up. For 23rd Century Fed (AoY) characters it is worth repeating the "Battle of Caleb IV" to gear up your ship. Every time my AoY character ranked up I repeated the "Battle of Caleb IV" to get upgraded gear for my ships. For other characters, missions like "Everything Old Is New" is a decent way to get Retrofit Phaser Banks to gear up your ship. Just look through the story mission link provided above to see what gear are rewarded. Remember that the mark level of the gear depends on your character's rank, but they generally max out at Mk XI.

Unfortunately, STO is too complex for one person to write a walkthrough or general guide because the game is too complex and no one guide will apply to everyone. On top of that there would be an overwhelming amount of information. The best thing to do is ask questions and grow your knowledge at a pace that you can absorb it.
