Ok clearly..
GWF is best for not using encounters.
HR is best for non at will (with cw second)
Alot of the the challenges are not class specific (donating to coffer) and such doesnt matter.
But I cant get the 50% under health one done.. every time.. random life steal proc , even on my under 10% toons.. i fail every time.
I think the only way to do it is a low level gwf alt or something. where you wont die .. but you can maintain it under 50% .
Maybe a gf or pally would work as well.

DC could compete with the HR for best for encounter only because divine encounters count. That's six encounters right off the bat and since healer dcs stack recovery they usually are ready for another rotation as soon the divine casting is done. Also DC is less likely to kill the targets which means less time spent finding mobs
Here's my list but note that I went with generic classes not specific builds like conq GF or righteous DC
Kill 2 foes without using any healing potions - GWF since they are top dps
Kill 5 foes within 1 minute - GWF since they are top dps
Donate to your Stronghold’s coffers within 3 minutes. Only appears on Stronghold maps. - anyone willing to dedicate 5 minutes to this very stupid challenge
Kill 10 foes without using any Daily Powers - GWF since they are top dps
Kill 10 foes without using any Encounter Powers - HR because of having twice as many encounters as everyone else
Survive 30 seconds of combat while under half health - DC (drop the health, then drop down hallowed ground, astral shield, and dodges to last 30 seconds) or GF (take damage and lift shield)
Use Daily Powers 3 times in 5 minutes - DC thanks to DC artifact and high recovery
Use Encounter Powers 10 times in combat - HR (has more encouters) or DC (divine encounters count as encounters and high recovery)
Complete 1 Heroic Encounter within 10 minutes - GWF since they are top dps
Use 3 skill nodes within 10 minutes - TR i guess, they can drop agro via stealth to get to nodes faster

Dismiss your companion (or use augment) so the monsters won't take any damage. Also remove any passive damage powers (e.g. OP Aura). Find some mobs, let them get you below 50%. Kill all but 1 mob. Let that last mob hit you (or not) while you do nothing to fight back. One mob shouldn't do enough to kill you (if so, drink a potion or run/dodge a little). Once the 30 sec are up and your gift appears, kill the single mob, heal up, be on your way.
You just have to be "in combat"... not actually attacking. If you're not attacking, no lifesteal. If they aren't hitting you, no boon heal procs. I find that portals work well for this (in dread ring / thayan magical shelter) or any big mob that is easy to dodge all the attacks (iron golem / green dragon cult / magma brute / etc). Avoid doing this with mobs that have high burst or hard to block / dodge attacks.
Taking off gear to make a challenge "easy" is just silly...
1. take off my SH lifesteal boon.
2. beat up the mob to lower their HP but not killing them.
3. Let them beat me up to below 50% and hope my other lifesteal/healing do not trigger. (This part is more time consuming).
4. Use dagger to finish them up.

But then again, I'm not trying this in Well of Dragons or something I aim more for Drowned / Hotenow / Fiery / Reclamation, where the biggest challenge is actually drawing enough mobs so that their damage will outweigh your passive regen (assuming you have 1 piece of Dusk gear, which is easy enough to obtain these days with Genie's Dusky Footwear flooding the AH for relatively cheap).

GWF for under 50% health go for spiders or imps, just stand there like a lemon till you are under 50% then Avalanche.
My GF was a bit trickier because he has a higher LS, I had to remove his rings and armour and did a minor HE, waited for them to knock me down to below 50% then use encounters, if my LS proc'd I'd wait till my health was low before using another encounter.
on my OP I find all of them (minus the Heroic and skill node ones) pretty easy. But when it comes to the below 50% ones I make sure I use Valorous Strike (since oath strike heals) and that none of my class features are of the healing variety. Then I dismiss my companion and just let a pack of mobs pounds on me and use Sanctuary to heal me up (since it's slow and easily controlled) if I happen to get into trouble.
If you're main objective is to farm the challenges, or just get the quick 5 for the daily quest, then I would stick to an area like the Drowned Coast for its plentiful and easily reaches mobs and for the fact that they can all (for the most part) be easily dispatched for the challenges requiring kills. The area seems to be one of the best places, IMO, for such a task.
For staying alive under 50% for 30 seconds, take out a tank companion. Let it hit under 50%. Don't hit anyone. Hence, no LS. After you are under 50%, start to run around and hope the mob go to your pet. For GWF, if the HP is low, bring up unstoppable to get more temp HP. For GF, you can drink a potion.
For killing 5 under 50%, it is easier but you may need all the time. Take out a tank companion. Unlike the above, you fails only if you run out of time. Lower some foe's HP. When your HP is low, throw dagger to kill something and hope LS does not trigger. If your HP is above 50%, let them hit you to down under. You just need to kill 5 independently when your HP is under 50%.
