I thought she was annoying and had to skip her dialogue half way through the episode. As a certain dinosaur theme park director once said, she suffers from a "deplorable excess of personality".

Melani removed non-orions from command positions, from her part of the Syndicate. They still employ other species, it would completely wreck them to expel non-orions as they would lose majority of the field agents, who would then just form their own crime organizations.
There are also other Syndicate cells who don't support D'ian and her schemes.

Not mention there would countless ex-members with grudge against D'ian who might now give a tip to the authorities (without revealing who did it obviously) about any illegal activities and it would be a lot harder to infiltrate organizations like Starfleet if you were working with just 1 species and if Ensign Tendi is any indication Orions who would be willing to join Starfleet normally wouldn't be that friendly with the Syndicate.
As a non-US person, I usually don't care about accents because I don't really recognize them as accents in the first place.
The fact I absolutely noticed this one and felt it was wrong was not a good sign.
There is no one southern accent. There are literally dozens, if not hundreds of regional variants. I'm not saying this one is accurate, but I don't think anyone can, either, unless they are a linguist.
I don't know the voice actor but she honestly sounded to me like somebody from southern California trying way too hard to sound like Texas. I mean, I like the flavor, but she overdid it.
For reference, I'm from central North Carolina. I've got a bit of the accent myself.

Something about the character design reminded me of "Doctor" Eric Cooper and I was instantly creeped out. (And after going to the wiki to look at images of Eric Cooper it's not a very strong resemblance, but somehow it's there.) The accent is also annoying, but just her basic appearance is disturbing. I was expecting her to also be an undine, I guess. So I personally do not want to see any more of her. It's not like there would be much difficulty in creating more characters for future missions.

It is a Tennessee mountain accent and those do vary a bit from area to area, and are completely different from the lowland accent of the state. While I am not an expert I used to have a friend with a mountain accent from that region , and Wilkins's is self-consistent and not bad from what I remember. It probably is a little exaggerated (or my friend's accent was slightly diluted by the local one), but that is something people with distinctive regional accents sometimes do on purpose around outsiders so it is reasonable.
The worst accent in the episode is the one the "Witch" has, that one is all over the place, she sometimes pronounces the same phonemes differently from word to word, often in the same sentence.
