I'm not sure we actually know enough about the First Federation to base any kind of missions around them
That's not necessarily a bad thing. Since the First Federation is a part of the current Federation, this could open up a lot of new possibilities for non-combat missions. For the devs, this is where they could get creative and beable to put their proverbial stamp on the Star Trek universe with out the restraints of canon. Because so little is known, they could be free to populate this space with Federation friendly species that are uniquely their own creations without the limitations of canon.
I've seen a thread some time ago proposing a series of civilian type missions prior to joining starfleet. After progressing through a series of missions and gaining experience in one specialty or another that will ultimately determine which class of officer you will be when you join Starfleet, go through the academy, and end up as an ensign thrust into Command of their own ship (where the current game begins).
An idea like this could work by expanding the Alpha Quadrant by adding a sector or two west of Apha Centari and Iota Pavonis sectors. Sol could be the point in space that connects the two sectors together. Since the First Federation is deep in Federation space, I would suspect that it would be one of the friendlier areas of space, well away from the frontlines, and a good area to have these preface, non-combat, diplomacy, civilian missions.
All goovyness aside, what did the OP mean with 'the first federation'? Are you talking about the federation in the time of jonathan archer or are you talking about finding some sort of 13th colony:P? What is this entire thread about for that matter?
