Steam: Right-click on Champions Online in your library and open Properties. In the Local Files tab, click on Verify Integrity of Game Cache and allow this to complete.

Thanks for the tip, unfortunately it didn't work for me. I'm getting the same error you were. When I verify the integrity through Steam, it says that two files would not validate and it would reacquire them. Afterward, when I verify the integrity, it says it worked.
When I lauch the game, it tries to connect to the launcher twice, then fails with the same error. I go to Steam and verify integrity -- and it says two files failed to validate. Attempted multiple times, all with the same result.
I checked the properties on the Champions Online Launcher and ran a compatibility test. As far as I could see, it didn't change any settings (was already set to Windows 8 and remained set to Windows 8), but the game worked after running the test.
